Complement fixation (CF) is performed first as the quantitative test. In the event of insufficient volume or equivocal or anticomplementary results by complement fixation methods, a quantitative immunodiffusion will be performed to determine the titer.
If the CF test is positive, residual specimen is frozen by us for later simultaneous testing with a new specimen to control for variability in the CF test (PRVEII). Residual specimen is saved for approximately 4-6 months.

The previously received specimen positive for coccidioidal IgG by complement fixation is retested as an internal control to account for natural variation in serology testing. Both results are released to help monitor subtle changes in patient titers. Residual specimen is saved for approximately 4-6 months.
